Kara Zor-El was sent to Earth from Krypton at age 13 by her parents Zor-El and Alura. Krypton was exploding, and Kara's parents sent Kara in a spaceship to Earth behind her cousin. Kara was destined to protect her baby cousin Kal-El, but her spaceship was diverted and forced into the Phantom Zone, where it remained for 24 years. By the time the spaceship landed on Earth, Kal-El had grown up to be Superman. The series begins eleven years later when Kara, 24, is learning to embrace her powers and has adopted the alias Supergirl.

In the first season, Kara is forced to reveal her powers and becomes the protector of National City. Kara discovers that hundreds of the criminals her mother imprisoned are hiding on Earth, including her aunt Astra and Astra's husband, Non. Kara works with her adoptive sister Alex Danvers to fight these criminals, along with the green Martian J'onn J'onzz, her cousin James Olsen's friend, and the technological genius Winn Schott.

In the second season, Kara and her allies grapple with disputes between Earth's native population and the extraterrestrial community, and investigate the shadowy organization Project Cadmus, devised by Lillian Luthor, mother of Lex Luthor. At the same time, Kara befriends Lillian's stepdaughter Lena Luthor, the new CEO of LuthorCorp, and struggles with romantic feelings over the recent arrival on Earth of Mon-El, a surviving prince from the neighboring planet of Krypton. Daxam, whose parents want to claim it. James becomes the masked vigilante Guardian; Alex begins dating Maggie Sawyer; and J'onn befriends a younger Martian, M'gann, of the white Martian race who killed his people
